چکیده
Human pluripotent stem cell (hPSCs) derived-pancreatic islets (hSC-islets) are good candidates for replacement therapy patients with diabetes as substitutes deceased donor-derived islets, because they and have infinite proliferation potential. Grafted hSC-islets ameliorate hyperglycemia in diabetic mice; however, several weeks needed to normalize the hyperglycemia. These data suggest require maturation, but their maturation process vivo is not yet fully understood. In this study, we utilized two kinds of streptozotocin (STZ)-induced model mice by changing administration timing order examine time course effects on maturation. We found no immune-compromised when had been transplanted under kidney capsules advance, STZ was administered 4 after transplantation. Of note, blood glucose levels those were stably maintained 100 mg/dL 10 transplantation; lower than mouse glycemic set point (120–150 mg/dL), suggesting that control human points. confirmed gene expression markers pancreatic beta cells tended upregulate during Periodical histological analysis revealed revascularization observed early 1 week transplantation, reinnervation grafted detected at all, even 15 conclusion, our need least mature, a index evaluating ultimate maturity vivo.

Maturation of Human Embryonic Stem Cell–Derived Pancreatic Progenitors Into Functional Islets Capable of Treating Pre-existing Diabetes in Mice
Diabetes is a chronic debilitating disease that results from insufficient production of insulin from pancreatic β-cells. Islet cell replacement can effectively treat diabetes but is currently severely limited by the reliance upon cadaveric donor tissue. We have developed a protocol to efficiently differentiate commercially available human embryonic stem cells (hESCs) in vitro into a highly enri...
متن کاملCo-Transplantation of VEGF-Expressing Human Embryonic Stem Cell Derived Mesenchymal Stem Cells to Enhance Islet Revascularization in Diabetic Nude Mice
Background: Pancreatic islet transplantation has emerged as a promising treatment for type I diabetes. However, its efficacy is severely hampered due to poor islet engraftment and revascularization, which have been resulted to partially loss of transplanted islets. It has been shown that local delivery of vascular endothelial growth factor (VEGF) could accelerate transplanted islet revasculari...

in the present study, the effect of e-64 at different concentrations (0.5, 1, 5 and 10 µm) added to (1) the ivm medium on oocyte nuclear maturation and developmental competence of ovine oocytes, and (2) to the ivc medium on embryonic development of ovine embryos were investigated.
Grafted human-induced pluripotent stem-cell-derived neurospheres promote motor functional recovery after spinal cord injury in mice.
Once their safety is confirmed, human-induced pluripotent stem cells (hiPSCs), which do not entail ethical concerns, may become a preferred cell source for regenerative medicine. Here, we investigated the therapeutic potential of transplanting hiPSC-derived neurospheres (hiPSC-NSs) into nonobese diabetic (NOD)-severe combined immunodeficient (SCID) mice to treat spinal cord injury (SCI). For th...
